Function of DQ2 and DQ8 as HLA susceptibility molecules in celiac disease.

چکیده
Celiac disease (CD) is a malabsorptive disorder of the small intestine. The disease is a unique model for studies of HLA-associated diseases since (a) the disease shows a stringent HLA association; (b) the disease-inducing agent (wheat gluten and related antigens from other cereals) is known; (c) activated, gluten-specific CD4 + T cells in the lamina propria of the small intestine most likely play an important immunopathogenic role; and (d) the target organ is accessible for obtaining biopsy specimens. Here we report studies on the mechanism behind the HLA association.

Gluten-specific T cells cross-react between HLA-DQ8 and the HLA-DQ2α/DQ8β transdimer.
Because susceptibility to celiac disease is associated strongly with HLA-DQ2 (DQA1*05/DQB1*02) and weakly with HLA-DQ8 (DQA1*03/DQB1*03), a subset of patients carries both HLA-DQ2 and HLA-DQ8. As a result, these patients may express two types of mixed HLA-DQ2/8 transdimers (encoded by DQA1*05/DQB1*03 and DQA1*03/DQB1*02) in addition to HLA-DQ2 and HLA-DQ8. Using T cells from a celiac disease pa...
متن کاملCystic Fibrosis and Celiac Disease: Mere Coincidence?
Aim: An increased prevalence of celiac disease (CD) in cystic fibrosis (CF) has been discussed. The aim of this survey was to determine the prevalence of a genetic predisposition to CD in CF-patients compared to the general population. Methods: Celiac serology and HLA-DQ2/ -DQ8 screening was performed in 190 CF-patients at the CF-Center Cologne, Germany. Informed consent was obtained from all p...
متن کاملHLA-DQ2 and -DQ8 signatures of gluten T cell epitopes in celiac disease.
Celiac disease is associated with HLA-DQ2 and, to a lesser extent, HLA-DQ8. Type 1 diabetes is associated with the same DQ molecules in the opposite order and with possible involvement of trans-encoded DQ heterodimers. T cells that are reactive with gluten peptides deamidated by transglutaminase 2 and invariably restricted by DQ2 or DQ8 can be isolated from celiac lesions. We used intestinal T ...
متن کاملRisk of Pediatric Celiac Disease According to HLA Haplotype and Country
This multi-centric cohort study [1] from four countries followed a group of infants with the HLA haplotype DR3– DQ2 or DR4–DQ8, from birth through the first few years of life; seeking the appearance of antibodies to tissue transglutaminase (tTG) (labeled as celiac disease autoimmunity), and development of celiac disease. This was part of a larger study evaluating the development of type 1 diabe...
متن کاملThe Prevalence of Hla Dq2 and Dq8 in Patients with Celiac Disease, in Family and in General Population
BACKGROUND Celiac disease is an enteropathy characterized by gluten sensitivity and broad clinical aspect. Has a multifactorial cause and depends on genetic, immunological and environmental factors for its development. The genetic influence is given mostly by the human leukocyte antigens HLA DQ2 and DQ8. AIM To evaluate the prevalence of human leukocyte antigens DQ2 and DQ8 in three different...
متن کاملThe incidence of HLA-DQ2/DQ8 in Turkish children with celiac disease and a comparison of the geographical distribution of HLA-DQ
Introduction Celiac disease (CD) is an auto-immune enteropathy that occurs in genetically pre-disposed people as a result of the consumption of gluten-containing foods. Aim To identify the incidence of HLA-DQ2 and HLA-DQ8 observed in children with CD. Material and methods In this study, we focused on children ranging in age from 2 to 18 years and diagnosed with celiac disease. In our patien...
